The origin and early evolution of membrane channels   总被引:3,自引:0,他引:3  
The origin and early evolution of ion channels are considered from the point of view that the transmembrane segments of membrane proteins are structurally quite simple and do not require specific sequences to fold. We argue that the transport of solute species, especially ions, required an early evolution of efficient transport mechanisms, and that the emergence of simple ion channels was protobiologically plausible. We also argue that, despite their simple structure, such channels could possess properties that, at the first sight, appear to require markedly greater complexity. These properties can be subtly modulated by local modifications to the sequence rather than global changes in molecular architecture. In order to address the evolution and development of ion channels, we focus on identifying those protein domains that are commonly associated with ion channel proteins and are conserved throughout the three main domains of life (Eukarya, Bacteria, and Archaea). We discuss the potassium-sodium-calcium superfamily of voltage-gated ion channels, mechanosensitive channels, porins, and ABC-transporters and argue that these families of membrane channels have sufficiently universal architectures that they can readily adapt to the diverse functional demands arising during evolution.  相似文献

The microbial diversity of Kali chimney plumes, part of a hydrothermal vent field in the Rodriguez Triple Junction, Indian Ocean (depth approximately 2,240 m), was examined in an attempt to discover "extremotolerant" microorganisms that have evolved unique resistance capabilities to this harsh environment. Water and sediment samples were collected from the vent and from sediments located at various distances (2-20 m) away from and surrounding the chimney. Samples were screened for hypertolerant microbes that are able to withstand multiple stresses. A total of 46 isolates were selected for exposure to a number of perturbations, such as heat shock, desiccation, H(2)O(2), and ultraviolet (UV) and gamma-irradiation. The survival of Psychrobacter sp. L0S3S-03b following exposure to >1,000 J/m(2) UV(254) radiation was particularly intriguing amid a background of varying levels of resistance. Vegetative cells of this non-spore-forming microbe not only survived all of the treatments, but also exhibited a 90% lethal dose of 30 s when exposed to simulated martian UV radiation and a 100% lethal dose of 2 min when exposed to full spectrum UV, which is comparable to findings for bacterial endospores.  相似文献

The Mars Program Plan includes an integrated and coordinated set of future candidate missions and investigations that meet fundamental science objectives of NASA and the Mars Exploration Program (MEP). At the time this paper was written, these possible future missions are planned in a manner consistent with a projected budget profile for the Mars Program in the next decade (2007-2016). As with all future missions, the funding profile depends on a number of factors that include the exact cost of each mission as well as potential changes to the overall NASA budget. In the current version of the Mars Program Plan, the Astrobiology Field Laboratory (AFL) exists as a candidate project to determine whether there were (or are) habitable zones and life, and how the development of these zones may be related to the overall evolution of the planet. The AFL concept is a surface exploration mission equipped with a major in situ laboratory capable of making significant advancements toward the Mars Program's life-related scientific goals and the overarching Vision for Space Exploration. We have developed several concepts for the AFL that fit within known budget and engineering constraints projected for the 2016 and 2018 Mars mission launch opportunities. The AFL mission architecture proposed here assumes maximum heritage from the 2009 Mars Science Laboratory (MSL). Candidate payload elements for this concept were identified from a set of recommendations put forth by the Astrobiology Field Laboratory Science Steering Group (AFL SSG) in 2004, for the express purpose of identifying overall rover mass and power requirements for such a mission. The conceptual payload includes a Precision Sample Handling and Processing System that would replace and augment the functionality and capabilities provided by the Sample Acquisition Sample Processing and Handling system that is currently part of the 2009 MSL platform.  相似文献

The relative abundance of the protein amino acids has been previously investigated as a potential marker for biogenicity in meteoritic samples. However, these investigations were executed without a quantitative metric to evaluate distribution variations, and they did not account for the possibility of interdisciplinary systematic error arising from inter-laboratory differences in extraction and detection techniques. Principal component analysis (PCA), hierarchical cluster analysis (HCA), and stochastic probabilistic artificial neural networks (ANNs) were used to compare the distributions for nine protein amino acids previously reported for the Murchison carbonaceous chondrite, Mars meteorites (ALH84001, Nakhla, and EETA79001), prebiotic synthesis experiments, and terrestrial biota and sediments. These techniques allowed us (1) to identify a shift in terrestrial amino acid distributions secondary to diagenesis; (2) to detect differences in terrestrial distributions that may be systematic differences between extraction and analysis techniques in biological and geological laboratories; and (3) to determine that distributions in meteoritic samples appear more similar to prebiotic chemistry samples than they do to the terrestrial unaltered or diagenetic samples. Both diagenesis and putative interdisciplinary differences in analysis complicate interpretation of meteoritic amino acid distributions. We propose that the analysis of future samples from such diverse sources as meteoritic influx, sample return missions, and in situ exploration of Mars would be less ambiguous with adoption of standardized assay techniques, systematic inclusion of assay standards, and the use of a quantitative, probabilistic metric. We present here one such metric determined by sequential feature extraction and normalization (PCA), information-driven automated exploration of classification possibilities (HCA), and prediction of classification accuracy (ANNs).  相似文献

This paper shares an interesting and unique case study of knowledge capture by the National Aeronautics and Space Administration (NASA), an ongoing project to recapture and make available the lessons learned from the Apollo lunar landing project so that those working on future projects do not have to “reinvent the wheel”. NASA’s new Constellation program, the successor to the Space Shuttle program, proposes a return to the Moon using a new generation of vehicles. The Orion Crew Vehicle and the Altair Lunar Lander will use hardware, practices, and techniques descended and derived from Apollo, Shuttle, and the International Space Station. However, the new generation of engineers and managers who will be working with Orion and Altair are largely from the decades following Apollo, and are likely not well aware of what was developed in the 1960s. In 2006, a project at NASA’s Johnson Space Center was started to find pertinent Apollo-era documentation and gather it, format it, and present it using modern tools for today’s engineers and managers. This “Apollo Mission Familiarization for Constellation Personnel” project is accessible via the web from any NASA center for those interested in learning answers to the question “how did we do this during Apollo?”  相似文献

Since the early 1990s the USA has maintained export control sanctions against Chinese international commercial satellite launch services. In 1998 these sanctions were further strengthened, resulting in a de facto international embargo that is premised on ITAR export licenses. Since 1998 this de facto embargo has effectively prohibited China from launching Western commercial satellites of a sophisticated technological standard. Today, European commercial satellite manufacturers are positioned to fully benefit from ITAR-free technology investments, gaining access to Chinese launch services for the launch of commercial telecommunication satellites that are technologically comparable to US satellites. This article examines the policy implications for the USA in light of EU regulatory divergence and the impending return of China to the international commercial launch services market.  相似文献

The German Research Centre for Geosciences (GFZ) operates a GNSS water vapour tomography system using about 350 German GNSS stations. The GNSS data processing at the GFZ works in near real-time and provides zenith total delays, integrated water vapour and slant delay data operationally. This large data set of more than 50,000 slant delays per hour is used to reconstruct spatially resolved humidity fields by means of tomographic techniques. It can be expected that additional observations from the future Galileo system provide more information with improved quality. A simulation study covering 12 h at 14 July 2009 was therefore started to estimate the impact of GPS, Galileo and GLONASS data on the GNSS tomography. It is shown that the spatial coverage of the atmosphere with slant paths is highly improved by combining observations from two or three satellite systems. Equally important for a reliable tomographic reconstruction is the distribution of slant path intersections as they are required to locate the integrated delay information. The number of intersection points can be increased by a factor of 4 or 8 if two or three systems are combined and their distribution will cover larger regions of the atmosphere. The combined data sets can be used to increase the spatiotemporal resolution of the reconstructed humidity fields up to 30 km horizontally, 300 m vertically and 15 min. The reconstruction quality could not be improved considerably using the currently available techniques.  相似文献

The EPOXI Discovery Mission of Opportunity reused the Deep Impact flyby spacecraft to obtain spatially and temporally resolved visible photometric and moderate resolution near-infrared (NIR) spectroscopic observations of Earth. These remote observations provide a rigorous validation of whole-disk Earth model simulations used to better understand remotely detectable extrasolar planet characteristics. We have used these data to upgrade, correct, and validate the NASA Astrobiology Institute's Virtual Planetary Laboratory three-dimensional line-by-line, multiple-scattering spectral Earth model. This comprehensive model now includes specular reflectance from the ocean and explicitly includes atmospheric effects such as Rayleigh scattering, gas absorption, and temperature structure. We have used this model to generate spatially and temporally resolved synthetic spectra and images of Earth for the dates of EPOXI observation. Model parameters were varied to yield an optimum fit to the data. We found that a minimum spatial resolution of ~100 pixels on the visible disk, and four categories of water clouds, which were defined by using observed cloud positions and optical thicknesses, were needed to yield acceptable fits. The validated model provides a simultaneous fit to Earth's lightcurve, absolute brightness, and spectral data, with a root-mean-square (RMS) error of typically less than 3% for the multiwavelength lightcurves and residuals of ~10% for the absolute brightness throughout the visible and NIR spectral range. We have extended our validation into the mid-infrared by comparing the model to high spectral resolution observations of Earth from the Atmospheric Infrared Sounder, obtaining a fit with residuals of ~7% and brightness temperature errors of less than 1?K in the atmospheric window. For the purpose of understanding the observable characteristics of the distant Earth at arbitrary viewing geometry and observing cadence, our validated forward model can be used to simulate Earth's time-dependent brightness and spectral properties for wavelengths from the far ultraviolet to the far infrared. Two possible scenarios for the detection of extraterrestrial intelligence are presented and the issues they would raise discussed. The article then goes on to examine operational questions and reply policy in the event of contact being made. Questions of how such contact should be announced to the world--and by whom; who should send a reply; how it would be designed and what it would say, are explored. Creating a beacon actively to beam a message into the galaxy could provide a test case for international consultation and cooperation in responding to ETI.  相似文献
